Abstract

There are several vesicular drug delivery systems developed as controlled and targeted drug delivery systems. Many constraints of various drug delivery systems, such as problems of drug incorporation, leakage from the carrier, insufficient shelf life, can be avoided by the pharmacosome approach. Pharmacosomes are one of the novel vesicular drug delivery systems. They are colloidal dispersion of the drug covalently bound to the lipids. They provide an efficient method of delivering drugs to the target site. Pharmacosomes, like other vesicular systems, offer an excellent way to convey medicine directly to the site of infection. They reduce drug toxicity without causing any adverse side effects. Furthermore, they reduce the cost of treatment through increasing drug bioavailability, mainly in the case of poorly soluble medicines. This strategy as a drug delivery system no doubt assures a reliable, safe, targeted, and exact drug delivery mechanism. The current review summarizes the latest updates about pharmacosomes and mechanisms by which they enhance the drug bioavailability. Further, it comprises information regarding methods of preparation and characterization and application of the pharmacosome in drug delivery. The increasing number of recent research addressing the utilization of drug–phospholipid complexes for the enhancement of the drug's oral bioavailability proves how important this approach is for successful oral delivery.
